Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies. This sequence change replaces lysine with asparagine at codon 41 of the KCNE1 protein (p.Lys41Asn). The lysine residue is moderately conserved and there is a moderate physicochemical difference between lysine and asparagine. - |

The lysine at codon 41 is replaced by asparagine, an amino acid with similar properties. Based on data from ExAC, the C allele has an overall frequency less than 0.01% (1/106190), having been observed in 0.006% (1/164080) of South Asian alleles. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
