The p.P333R variant (also known as c.998C>G), located in coding exon 8 of the RUNX1 gene, results from a C to G substitution at nucleotide position 998. The proline at codon 333 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.

This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 333 of the RUNX1 protein (p.Pro333Arg). This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RUNX1-related conditions. ClinVar contains an entry for this variant (Variation ID: 572808).
