This sequence change replaces arginine with cysteine at codon 54 of the CRYAA protein (p.Arg54Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is not present in population databases (rs397515623, ExAC no frequency). This variant has been reported to segregate in two families affected with bilateral punctate lenticular opacities and congenital cataracts associated with microcornea (PMID: 17937925, 18587492). Both individuals who were heterozygous for the variant and individuals who were homozygous for the variant were affected. ClinVar contains an entry for this variant (Variation ID: 68457). Experimental studies have shown that this variant affects the structure and function of the alpha-crystallin protein (PMID: 22045060, 22347476, 22140512, 23379525, 25018622, 25694240, 28179137). For these reasons, this variant has been classified as Pathogenic. -
