PCNT (HGNC:16068): (pericentrin) The protein encoded by this gene binds to calmodulin and is expressed in the centrosome. It is an integral component of the pericentriolar material (PCM). The protein contains a series of coiled-coil domains and a highly conserved PCM targeting motif called the PACT domain near its C-terminus. The protein interacts with the microtubule nucleation component gamma-tubulin and is likely important to normal functioning of the centrosomes, cytoskeleton, and cell-cycle progression. Mutations in this gene cause Seckel syndrome-4 and microcephalic osteodysplastic primordial dwarfism type II. Two transcript variants encoding different isoforms have been found for this gene. The E1037X pathogenic variant in the PCNT gene has been reported previously in the homozygous state in individuals with MOPDII from consanguineous families (Rauch et al., 2008; Willems et al., 2010). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Fibroblasts from an individual homozygous for the E1037X variant demonstrated abnormal mitotic morphology, low-level mosaic variegated aneuploidy, and premature sister chromatid separation (Rauch et al., 2008). The E1037X variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret E1037X as a pathogenic variant. -
