This variant has not been reported in the literature in individuals affected with CHEK2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1762953). The p.C420* pathogenic mutation (also known as c.1260C>A), located in coding exon 11 of the CHEK2 gene, results from a C to A substitution at nucleotide position 1260. This changes the amino acid from a cysteine to a stop codon within coding exon 11. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
