In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with CHEK2-related conditions. This sequence change replaces histidine with arginine at codon 345 of the CHEK2 protein (p.His345Arg). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and arginine. This variant is not present in population databases (ExAC no frequency).
Hereditary cancer-predisposing syndrome Uncertain:1
The p.H345R variant (also known as c.1034A>G), located in coding exon 9 of the CHEK2 gene, results from an A to G substitution at nucleotide position 1034. The histidine at codon 345 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
