This sequence change replaces leucine, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 236 of the CHEK2 protein (p.Leu236Gln). This variant is present in population databases (no rsID available, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with CHEK2-related conditions. ClinVar contains an entry for this variant (Variation ID: 232155). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Leu236 amino acid residue in CHEK2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 25186627, 25318351, 30851065, 31206626, 37449874; internal data). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. The p.L236Q variant (also known as c.707T>A), located in coding exon 5 of the CHEK2 gene, results from a T to A substitution at nucleotide position 707. The leucine at codon 236 is replaced by glutamine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
