The c.684-4C>G intronic variant results from a C to G substitution 4 nucleotides upstream from coding exon 5 in the CHEK2 gene. This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing; however, direct evidence is insufficient at this time (Ambry internal data). In addition, the -4 position of the consensus splice acceptor site is tolerant to nucleotide substitution (Padgett RA Trends Genet. 2012 Apr 28;4:147-154). Based on the available evidence, the clinical significance of this variant remains unclear. -
