The p.Y136H variant (also known as c.406T>C), located in coding exon 2 of the CHEK2 gene, results from a T to C substitution at nucleotide position 406. The tyrosine at codon 136 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces tyrosine, which is neutral and polar, with histidine, which is basic and polar, at codon 136 of the CHEK2 protein (p.Tyr136His). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CHEK2-related conditions. ClinVar contains an entry for this variant (Variation ID: 460832).
