The p.R95Q variant (also known as c.284G>A), located in coding exon 1 of the CHEK2 gene, results from a G to A substitution at nucleotide position 284. The arginine at codon 95 is replaced by glutamine, an amino acid with highly similar properties. This alteration was observed in female breast cancer patients and at least one control across several studies (Momozawa Y et al. Nat Commun. 2018 10;9:4083; Weitzel JN et al. Cancer 2019 08;125:2829-2836; Dorling N Engl J Med. 2021 02;384:428-439). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
This missense variant replaces arginine with glutamine at codon 95 of the CHEK2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer (PMID: 30287823). This variant has also been identified in 5/251208 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 95 of the CHEK2 protein (p.Arg95Gln). This variant is present in population databases (rs750596499, gnomAD 0.006%). This missense change has been observed in individual(s) with breast cancer (PMID: 30287823). ClinVar contains an entry for this variant (Variation ID: 230628). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
