A complementation assay in human cells showed that this variant has intermediate impact on KAP1 phosphorylation and no impact on CHEK2 autophosphorylation (PMID: 37449874). This variant has not been reported in individuals affected with CHEK2-related disorders in the literature, but has been reported in an unaffected individual (PMID: 37449874). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.Q11P variant (also known as c.32A>C), located in coding exon 1 of the CHEK2 gene, results from an A to C substitution at nucleotide position 32. The glutamine at codon 11 is replaced by proline, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species.
