Genes affected
SOX10 (HGNC:11190): (SRY-box transcription factor 10) This gene encodes a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. The encoded protein may act as a transcriptional activator after forming a protein complex with other proteins. This protein acts as a nucleocytoplasmic shuttle protein and is important for neural crest and peripheral nervous system development. Mutations in this gene are associated with Waardenburg-Shah and Waardenburg-Hirschsprung disease. [provided by RefSeq, Jul 2008]
POLR2F (HGNC:9193): (RNA polymerase II, I and III subunit F) This gene encodes the sixth largest subunit of RNA polymerase II, the polymerase responsible for synthesizing messenger RNA in eukaryotes. In yeast, this polymerase subunit, in combination with at least two other subunits, forms a structure that stabilizes the transcribing polymerase on the DNA template. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2014]

Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Oct 22, 2022 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SOX10 protein function. ClinVar contains an entry for this variant (Variation ID: 517217). This variant has not been reported in the literature in individuals affected with SOX10-related conditions. This variant is present in population databases (rs748234376, gnomAD 0.02%). This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 258 of the SOX10 protein (p.Arg258Gln). - |
Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Dec 02, 2024 | Observed as an assumed de novo variant in patients with autism spectrum disorders in published literature; however, the reported clinical features are not consistent with the features typically observed in individuals with pathogenic variants in this gene (PMID: 31785789, 35982159, 35982160, 25363768); In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; This variant is associated with the following publications: (PMID: 31785789, 34011629, 35982160, 31133750, 35982159, 25363768, 37217689) - |

Uncertain significance, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Jul 25, 2017 | The p.Arg258Gln variant in SOX10 has not been reported in individuals with heari ng loss or Waardenburg syndrome, but has been identified in 8/34416 Latino chrom osomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute. org; dbSNP rs748234376). Although this variant has been seen in the general popu lation, its frequency is not high enough to rule out a pathogenic role. Computat ional prediction tools and conservation analysis suggest that the p.Arg258Gln va riant may impact the protein, though this information is not predictive enough t o determine pathogenicity. In summary, the clinical significance of the p.Arg258 Gln variant is uncertain. - |
SOX10-related disorder Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | PreventionGenetics, part of Exact Sciences | Mar 23, 2023 | The SOX10 c.773G>A variant is predicted to result in the amino acid substitution p.Arg258Gln. This variant was reported as de novo in a patient with autism spectrum disorder (Table S2, Iossifov et al 2014. PubMed ID: 25363768), and to our knowledge has not been reported in patients with a SOX10 specific disorder. This variant is reported in 0.023% of alleles in individuals of Latino descent in gnomAD representing 18 heterozygous individuals (http://gnomad.broadinstitute.org/variant/22-38370130-C-T), which is high for a disease causing variant in this gene. While we suspect this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. - |
