It is the largest mammalian selenoprotein, containing the rare amino acid selenocysteine (Sec). Sec is encoded by the UGA codon, which normally signals translation termination. The 3' UTRs of selenoprotein mRNAs contain a conserved stem-loop structure, designated the Sec insertion sequence (SECIS) element, that is necessary for the recognition of UGA as a Sec codon, rather than as a stop signal. This variant has not been reported in the literature in individuals with TUBGCP6-related conditions. This variant is present in population databases (rs765823423, ExAC 0.003%). This sequence change replaces phenylalanine with tyrosine at codon 1812 of the TUBGCP6 protein (p.Phe1812Tyr). The phenylalanine residue is highly conserved and there is a small physicochemical difference between phenylalanine and tyrosine. -
