The encoded protein couples this information to intracellular signaling pathways that modify parathyroid hormone secretion or renal cation handling, and thus this protein plays an essential role in maintaining mineral ion homeostasis. Two of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00012 in 251360 control chromosomes (gnomAD). The observed variant frequency is approximately 99 fold of the estimated maximal expected allele frequency for a pathogenic variant in CASR causing Autosomal Dominant Hypocalcemia phenotype (1.3e-06), strongly suggesting that the variant is benign. c.779A>G has been reported in the literature in one individual affected with cystic fibrosis (Gaitch_2016). The report does not provide unequivocal conclusions about association of the variant with Autosomal Dominant Hypocalcemia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 27086061). Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014 and classified this variant as uncertain significance (n=2) and likely benign (n=1).
