This endonuclease catalyzes the first step in RNA splicing which is the removal of introns. Mutations in this gene have been associated with pontocerebellar hypoplasia type 2. A pseudogene has been identified on chromosome 4. Multiple transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Feb 2009]

This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 196 of the TSEN2 protein (p.Gly196Val). This variant is present in population databases (rs143243952, gnomAD 0.09%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with TSEN2-related conditions. ClinVar contains an entry for this variant (Variation ID: 437062).
