The p.D555H variant (also known as c.1663G>C), located in coding exon 14 of the RAF1 gene, results from a G to C substitution at nucleotide position 1663. The aspartic acid at codon 555 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 555 of the RAF1 protein (p.Asp555His). This variant is present in population databases (rs371704292, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RAF1-related conditions. ClinVar contains an entry for this variant (Variation ID: 503546). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt RAF1 protein function with a positive predictive value of 95%.
