This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 161 of the RAB7A protein (p.Asn161Ser). This variant is present in population databases (no rsID available, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with RAB7A-related conditions. ClinVar contains an entry for this variant (Variation ID: 846870). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant disrupts the p.Asn161 amino acid residue in RAB7A.
