This variant disrupts the p.Lys296 amino acid residue in RHO. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 11139241, 21677794, 31087526). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. This missense change has been observed in individual(s) with autosomal dominant retinitis pigmentosa (PMID: 1765377). It has also been observed to segregate with disease in related individuals. This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 296 of the RHO protein (p.Lys296Glu). This variant is not present in population databases (gnomAD no frequency).
