The encoded protein may play a role in organelle biogenesis associated with melanosomes, platelet dense granules, and lysosomes. CP (HGNC:2295): (ceruloplasmin) The protein encoded by this gene is a metalloprotein that binds most of the copper in plasma and is involved in the peroxidation of Fe(II)transferrin to Fe(III) transferrin. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in HPS3 are known to be pathogenic (PMID: 11590544). This variant is present in population databases (rs281865095, gnomAD 0.0009%). Disruption of this splice site has been observed in individual(s) with Hermansky-Pudlak syndrome (PMID: 11590544). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as 2729+1G>C. ClinVar contains an entry for this variant (Variation ID: 4612). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
