OPA1 (HGNC:8140): (OPA1 mitochondrial dynamin like GTPase) The protein encoded by this gene is a nuclear-encoded mitochondrial protein with similarity to dynamin-related GTPases. The encoded protein localizes to the inner mitochondrial membrane and helps regulate mitochondrial stability and energy output. This protein also sequesters cytochrome c. Mutations in this gene have been associated with optic atrophy type 1, which is a dominantly inherited optic neuropathy resulting in progressive loss of visual acuity, leading in many cases to legal blindness. This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 290 of the OPA1 protein (p.Arg290Trp). This variant is present in population databases (rs780333963, gnomAD 0.002%). This missense change has been observed in individual(s) with OPA1-related conditions (PMID: 11440988). ClinVar contains an entry for this variant (Variation ID: 402217). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Arg290 amino acid residue in OPA1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 11017080, 11440988, 11810270, 22779427, 25564500). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
