A different variant at this position, c.-27C>A, has been observed in Lynch syndrome families as a haplotype with the variant MLH1 c.85G>T (PMID: 16083711, 21840485, 22878509, 24084575) and the c.-27C>A variant exhibited reduced promotor activity in a luciferase assay (PMID: 21840485, 22878509). This haplotype is described as pathogenic (ClinVar variation ID: 220516). The available evidence is insufficient to determine the role of this variant in disease conclusively.
