Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and all classified the variant as uncertain significance. The glutamine at codon 426 is replaced by leucine, an amino acid with dissimilar properties. This alteration was identified in multiple individuals diagnosed with colorectal cancer (Fanale D et al. Front Oncol, 2022 Feb;12:827822). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
