The proline at codon 603 is replaced by leucine, an amino acid with similar properties. This variant was detected in a pediatric patient with adrenocortical tumor (ACT) who was also found to carry a pathogenic TP53 germline mutation (Brondani VB et al. Cancers (Basel), 2020 Mar;12). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
