This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 1033 of the SCN5A protein (p.Gln1033Arg). This variant is present in population databases (rs199473641, gnomAD 0.0009%). This missense change has been observed in individuals with clinical features of SCN5A-related conditions (PMID: 17605181, 26412604, 26669661, 31737537, 34147702). ClinVar contains an entry for this variant (Variation ID: 67776). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The arginine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

This variant has also been observed in an individual with hearing loss and epilepsy who was not diagnosed with long QT syndrome (PMID 26412604), and in another individual affected with global developmental delay (PMID: 38539105). This variant has been identified in 1/248740 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
