This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 986 of the SCN5A protein (p.Arg986Gln). This variant is present in population databases (rs41313667, gnomAD 0.02%). This missense change has been observed in individual(s) with irritable bowel syndrome and/or lone atrial fibrillation, dilated cardiomyopathy (PMID: 15851227, 20129283, 26129877, 29167113, 37652022). ClinVar contains an entry for this variant (Variation ID: 67770). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. Experimental studies have shown that this missense change affects SCN5A function (PMID: 29167113).
