The p.Tyr205X variant in SCN5A has not been previously reported in the literatur e, and has been identified in 1/26626 South Asian chromosomes by the Genome Aggr egation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs765669597). It has been reported in ClinVar (Variation ID: 201437). This nonsense variant le ads to a premature termination codon at position 205, which is predicted to lead to a truncated or absent protein. However, this nonsense variant occurs in an a lternative transcript of the SCN5A gene that has been shown to only be expressed during fetal development (Murphy 2012). In summary, the clinical significance o f the p.Tyr205X variant is uncertain.
