SCN5A (HGNC:10593): (sodium voltage-gated channel alpha subunit 5) The protein encoded by this gene is an integral membrane protein and tetrodotoxin-resistant voltage-gated sodium channel subunit. This protein is found primarily in cardiac muscle and is responsible for the initial upstroke of the action potential in an electrocardiogram. Defects in this gene have been associated with long QT syndrome type 3 (LQT3), atrial fibrillation, cardiomyopathy, and Brugada syndrome 1, all autosomal dominant cardiac diseases. Although the c.612-1 G>C variant has not been reported as a pathogenic or benign to our knowledge, it is predicted to destroy the canonical splice acceptor site in intron 5 and may cause abnormal gene splicing. Additionally, the c.612-1 G>C variant is not observed in large population cohorts (Lek et al., 2016). Nonetheless, there is limited information about the clinical significance of variants on this alternate transcript. No splice site or loss-of-function variants in this transcript of the SCN5A gene have been reported in HGMD in association with disease (Stenson et al., 2014). - |
