Submissions by phenotype
Brugada syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jul 19, 2022 | This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 741 of the SCN10A protein (p.Leu741Arg). This variant is present in population databases (rs201342036, gnomAD 0.004%). This missense change has been observed in individual(s) with Brugada syndrome (PMID: 31292628). ClinVar contains an entry for this variant (Variation ID: 407744). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SCN10A protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
Cardiovascular phenotype Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Apr 17, 2023 | The p.L741R variant (also known as c.2222T>G), located in coding exon 14 of the SCN10A gene, results from a T to G substitution at nucleotide position 2222. The leucine at codon 741 is replaced by arginine, an amino acid with dissimilar properties. This variant was identified in one individual with Brugada syndrome (Monasky MM et al. Europace, 2019 10;21:1550-1558). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on available evidence to date, the clinical significance of this alteration remains unclear. - |
