The p.E306K variant (also known as c.916G>A), located in coding exon 10 of the BAP1 gene, results from a G to A substitution at nucleotide position 916. The glutamic acid at codon 306 is replaced by lysine, an amino acid with similar properties. This alteration was identified in 1/183 unselected patients with BAP1-related disease (Zauderer MG et al. J Thorac Oncol, 2019 11;14:1989-1994).This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces glutamic acid with lysine at codon 306 of the BAP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in an individual affected with BAP1-related disease (PMID: 31323388).
