The p.W202C variant (also known as c.606G>C), located in coding exon 8 of the BAP1 gene, results from a G to C substitution at nucleotide position 606. The tryptophan at codon 202 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid change has been observed in individuals with a personal and/or family history that is consistent with BAP1-related disease (Ambry internal data; Personal communication; De Rienzo A et al. Cancer Res, 2016 Jan;76:319-28). A close match alteration at this same codon (c.604T>C, p.W202R) has been observed in a family with BAP1-associated phenotype (Kittaneh M et al. J Transl Med 2018 Jul;16(1):194). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic. -
