This nucleotide position is highly conserved in available vertebrate species. This alteration has been observed in individuals with a personal and/or family history that is consistent with BAP1-related disease (Testa JR et al. Nat Genet, 2011 Aug;43:1022-5). In vivo mouse studies have shown that mice with this alteration are significantly predisposed to BAP1-associated tumors (Kadariya Y et al. Cancer Res, 2016 05;76:2836-44). RNA studies have demonstrated that this alteration results in abnormal splicing in the set of samples tested (Ambry internal data; Testa JR et al. Nat Genet, 2011 Aug;43:1022-5). In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. Based on the supporting evidence, this alteration is interpreted as a disease-causing mutation. - |
