This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 149 of the TNNC1 protein (p.Asp149Asn). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with dilated cardiomyopathy (PMID: 27532257, 30847666, 37652022; internal data). ClinVar contains an entry for this variant (Variation ID: 44683). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The c.445G>A (p.D149N) alteration is located in exon 5 (coding exon 5) of the TNNC1 gene. This alteration results from a G to A substitution at nucleotide position 445, causing the aspartic acid (D) at amino acid position 149 to be replaced by an asparagine (N).
