These family members tether calcineurin to alpha-actinin at the z-line of the sarcomere of cardiac and skeletal muscle cells, and thus they are important for calcineurin signaling. This variant results from an A to G substitution 3 bases upstream from the first translated codon. This variant was previously reported in the SNPDatabase as rs372377840. This variant was not reported in population-based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This nucleotide position is not well conserved in available vertebrate species, and G is the reference nucleotide in six species. Since supporting evidence for this variant is limited at this time, the clinical significance of this variant remains unclear. - |
