Genes affected
PROM1 (HGNC:9454): (prominin 1) This gene encodes a pentaspan transmembrane glycoprotein. The protein localizes to membrane protrusions and is often expressed on adult stem cells, where it is thought to function in maintaining stem cell properties by suppressing differentiation. Mutations in this gene have been shown to result in retinitis pigmentosa and Stargardt disease. Expression of this gene is also associated with several types of cancer. This gene is expressed from at least five alternative promoters that are expressed in a tissue-dependent manner. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2009]

This sequence change falls in intron 18 of the PROM1 gene. It does not directly change the encoded amino acid sequence of the PROM1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with autosomal recessive PROM1-related conditions (PMID: 26153215). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 190373). Studies have shown that this variant alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 26153215). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
