PROM1 (HGNC:9454): (prominin 1) This gene encodes a pentaspan transmembrane glycoprotein. The protein localizes to membrane protrusions and is often expressed on adult stem cells, where it is thought to function in maintaining stem cell properties by suppressing differentiation. Mutations in this gene have been shown to result in retinitis pigmentosa and Stargardt disease. Expression of this gene is also associated with several types of cancer. This gene is expressed from at least five alternative promoters that are expressed in a tissue-dependent manner. This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 202 of the PROM1 protein (p.Arg202Gly). This variant is present in population databases (rs140872693, gnomAD 0.09%). This missense change has been observed in individual(s) with clinical features of PROM1-related conditions (PMID: 26355662, 31129250, 37217489, 37734845). ClinVar contains an entry for this variant (Variation ID: 191189). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on PROM1 protein function.
