Genes affected
QDPR (HGNC:9752): (quinoid dihydropteridine reductase) This gene encodes the enzyme dihydropteridine reductase, which catalyzes the NADH-mediated reduction of quinonoid dihydrobiopterin. This enzyme is an essential component of the pterin-dependent aromatic amino acid hydroxylating systems. Mutations in this gene resulting in QDPR deficiency include aberrant splicing, amino acid substitutions, insertions, or premature terminations. Dihydropteridine reductase deficiency presents as atypical phenylketonuria due to insufficient production of biopterin, a cofactor for phenylalanine hydroxylase. [provided by RefSeq, Jul 2008]

Dihydropteridine reductase deficiency Pathogenic:1
Likely pathogenic, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Jul 11, 2017 | The p.His158Tyr (NM_000320.2 c.472C>T) variant in QDPR has been reported in at l east 3 homozygous individuals with dihydropteridine reductase deficiency (Smooke r 1993, Dianzani 1998, and Foroozani 2015). This variant has been identified 1/6 5,896 of European chromosomes by the Exome Aggregation Consortium (ExAC, http:// exac.broadinstitute.org), and this frequency is low enough to be consistent with a recessive carrier frequency. Computational prediction tools and conservation analysis suggest that the p.His158Tyr variant may impact the protein, though thi s information is not predictive enough to determine pathogenicity. In summary, a lthough additional studies are required to fully establish its clinical signific ance, the p.His158Tyr variant is likely pathogenic for dihydropteridine reductas e deficiency in an autosomal recessive manner based upon biallelic observations in affected individuals. - |
