F11 (HGNC:3529): (coagulation factor XI) This gene encodes coagulation factor XI of the blood coagulation cascade. This protein is present in plasma as a zymogen, which is a unique plasma coagulation enzyme because it exists as a homodimer consisting of two identical polypeptide chains linked by disulfide bonds. During activation of the plasma factor XI, an internal peptide bond is cleaved by factor XIIa (or XII) in each of the two chains, resulting in activated factor XIa, a serine protease composed of two heavy and two light chains held together by disulfide bonds. This activated plasma factor XI triggers the middle phase of the intrisic pathway of blood coagulation by activating factor IX. Defects in this factor lead to Rosenthal syndrome, a blood coagulation abnormality. [provided by RefSeq, Jul 2008]

Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251482 control chromosomes (gnomAD). c.1207G>A has been reported in the literature in a homozygous individual affected with Hereditary factor XI deficiency disease (Dai_2011), as well as 2 heterozygous individuals reported to have a coagulation disorder (Downes_2019). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, finding that the variant resulted in sharply decreased extracellular secretion (5.7% of wild type antigen level), as well as inhibiting secretion of heterodimers of the mutant and wild type protein (Dai_2011), suggesting a possible dominant negative effect. The following publications have been ascertained in the context of this evaluation (PMID: 21457405, 31064749). ClinVar contains an entry for this variant (Variation ID: 68173).
