DOK7 (HGNC:26594): (docking protein 7) The protein encoded by this gene is essential for neuromuscular synaptogenesis. The protein functions in aneural activation of muscle-specific receptor kinase, which is required for postsynaptic differentiation, and in the subsequent clustering of the acetylcholine receptor in myotubes. This protein can also induce autophosphorylation of muscle-specific receptor kinase. Mutations in this gene are a cause of familial limb-girdle myasthenia autosomal recessive, which is also known as congenital myasthenic syndrome type 1B. A variant of uncertain significance has been identified in the DOK7 gene. The A482V variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The A482Vvariant is observed in 5/6046 (0.1%) alleles from individuals of Latino background (Lek et al., 2016). The A482V variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is not conserved, and in silico predicts this variant likely does not alter the protein structure/function. Based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant. -

This variant is present in population databases (rs767980190, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with DOK7-related conditions.
