The protein is a phosphatidylinositol (3,4,5)-trisphosphate-dependent Arf6 GAP that binds RhoA-GTP, but it lacks the predicted catalytic arginine in the RHO-GAP domain and does not have RHO-GAP activity. The c.5014G>A (p.D1672N) alteration is located in exon 33 (coding exon 32) of the ARAP2 gene. This alteration results from a G to A substitution at nucleotide position 5014, causing the aspartic acid (D) at amino acid position 1672 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
