The protein is a phosphatidylinositol (3,4,5)-trisphosphate-dependent Arf6 GAP that binds RhoA-GTP, but it lacks the predicted catalytic arginine in the RHO-GAP domain and does not have RHO-GAP activity. The c.4891C>T (p.R1631W) alteration is located in exon 33 (coding exon 32) of the ARAP2 gene. This alteration results from a C to T substitution at nucleotide position 4891, causing the arginine (R) at amino acid position 1631 to be replaced by a tryptophan (W). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
