The c.371C>G (p.T124R) alteration is located in exon 3 (coding exon 3) of the PIGG gene. This alteration results from a C to G substitution at nucleotide position 371, causing the threonine (T) at amino acid position 124 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces threonine with arginine at codon 124 of the PIGG protein (p.Thr124Arg). The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and arginine. This variant is present in population databases (rs370385328, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with PIGG-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
