The c.1900G>C (p.D634H) alteration is located in exon 9 (coding exon 9) of the PIGG gene. This alteration results from a G to C substitution at nucleotide position 1900, causing the aspartic acid (D) at amino acid position 634 to be replaced by a histidine (H). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces aspartic acid with histidine at codon 634 of the PIGG protein (p.Asp634His). The aspartic acid residue is weakly conserved and there is a moderate physicochemical difference between aspartic acid and histidine. This variant is present in population databases (rs145542247, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with PIGG-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
