Submissions by phenotype
Gastrointestinal stromal tumor Uncertain:1
This variant has not been reported in the literature in individuals affected with PDGFRA-related conditions. ClinVar contains an entry for this variant (Variation ID: 1019626). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces aspartic acid, which is acidic and polar, with tyrosine, which is neutral and polar, at codon 756 of the PDGFRA protein (p.Asp756Tyr). -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.D756Y variant (also known as c.2266G>T), located in coding exon 15 of the PDGFRA gene, results from a G to T substitution at nucleotide position 2266. The aspartic acid at codon 756 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
