Mutations in this gene are associated with Wolfram syndrome, also called DIDMOAD (Diabetes Insipidus, Diabetes Mellitus, Optic Atrophy, and Deafness), an autosomal recessive disorder. The disease affects the brain and central nervous system. Mutations in this gene can also cause autosomal dominant deafness 6 (DFNA6), also known as DFNA14 or DFNA38. Ser790Trp in exon 8 of WFS1: This variant has not been identified in individuals with hearing loss, but has been identified in 0.09% (4/4296) of African America n chromosomes from a broad population by the NHLBI Exome Sequencing Project (htt p://evs.gs.washington.edu/EVS). Although this variant has been seen in the gener al population, its frequency is not high enough to rule out a pathogenic role. H owever, the Ser790 residue is poorly conserved in mammals as well as found as a Trp residue in several fish species. As such, this variant is not likely to have clinical significance.
