Action myoclonus-renal failure syndrome Uncertain:2
This variant has not been reported in the literature but is present in the Genome Aggregation Database (Highest reported MAF 0.02% (3/15280) (https://gnomad.broadinstitute.org/variant/4-76163352-C-T?dataset=gnomad_r3). This variant is present in ClinVar (Variation ID:206705). This variant amino acid Glutamine (Gln) is present in several species including multiple mammals and is not well conserved among evolutionarily distant species; this suggests that this variant may not impact the protein. Additional computational prediction tools do not suggest an impact. In summary, data on this variant is insufficient for disease classification. Therefore, the clinical significance of this variant is uncertain.
not provided Uncertain:2
In silico analysis indicates that this missense variant does not alter protein structure/function; Has not been previously published as pathogenic or benign to our knowledge; This variant is associated with the following publications: (PMID: 28492532)
Inborn genetic diseases Uncertain:1
The p.R424Q variant (also known as c.1271G>A), located in coding exon 11 of the SCARB2 gene, results from a G to A substitution at nucleotide position 1271. The arginine at codon 424 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and glutamine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
Progressive myoclonic epilepsy Uncertain:1
This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 424 of the SCARB2 protein (p.Arg424Gln). This variant is present in population databases (rs751827409, gnomAD 0.2%). This variant has not been reported in the literature in individuals affected with SCARB2-related conditions. ClinVar contains an entry for this variant (Variation ID: 206705). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SCARB2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
