J Mol Diagn. 2013 Jan;15(1):31-43; Ambry internal data). Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 251116 control chromosomes. c.221-2A>G has been reported in the literature in individuals affected with Familial Adenomatous Polyposis, Colorectal Cancer and Attenuated familial adenomatous polyposis (example, Beaton_2008, Kerr_2013,Svensson_2022). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
