The p.M271L variant (also known as c.811A>T), located in coding exon 7 of the APC gene, results from an A to T substitution at nucleotide position 811. The methionine at codon 271 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

The p.Met271Leu variant in APC has not been reported in the literature in indivi duals with APC-associated polyposis but has been reported by other clinical labo ratories in ClinVar (Variation ID: 482436). This variant was absent from large p opulation studies. Computational prediction tools and conservation analysis sugg est that the p.Met271Leu variant may not impact the protein.
