It is also involved in other processes including cell migration and adhesion, transcriptional activation, and apoptosis. Defects in this gene cause familial adenomatous polyposis (FAP), an autosomal dominant pre-malignant disease that usually progresses to malignancy. The c.1239dupA pathogenic mutation, located in coding exon 9 of the APC gene, results from a duplication of A at nucleotide position 1239, causing a translational frameshift with a predicted alternate stop codon (p.R414Tfs*5). This mutation has been previously reported in families with classic FAP presentation, as well as extracolonic tumors, such as hepatocellular, pancreatic and duodenal cancers (Kerr SE et al. J Mol Diagn 2013 Jan; 15(1):31-43; Andresen PA et al. J. Cancer Res. Clin. Oncol. 2009 Oct; 135(10):1463-70). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
