The p.Q445* pathogenic mutation (also known as c.1333C>T), located in coding exon 10 of the APC gene, results from a C to T substitution at nucleotide position 1333. This changes the amino acid from a glutamine to a stop codon within coding exon 10. This mutation (designated p.Gln445X, c.1335C>T) was identified in one of 107 patients with FAP-like phenotype, with approximately 100 adenomas being noted throughout this individual's colon at age 37 (Filipe B et al. Clin. Genet. 2009 Sep;76:242-55). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
