However, these predictions have yet to be functionally assessed. The variant of interest was not observed in 120344 control chromosomes (ExAC). The variant of interest has not been, to our knowledge, reported in affected individuals via publications, although a clinical diagnostic laboratory cites the variant with a classification of "uncertain significance." The p.L456F variant (also known as c.1366C>T), located in coding exon 10 of the APC gene, results from a C to T substitution at nucleotide position 1366. The leucine at codon 456 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
